The Role of Orexin

NT1 is now recognized as a highly selective, autoimmune-mediated destruction of the orexin-producing neurons in the lateral hypothalamus. Only 23.1% of physicians in a recent survey correctly identified low levels of orexin as characteristic of NT1.

Genetic Factor: The primary genetic risk factor is the HLA-DQB1*06:02 allele, present in over 98% of NT1 cases, allowing presentation of orexin-derived peptides to T-cells.
